Description


Androstane-diones are a pair of isomeric steroids which differ at the 5-position. They are distinguished as 5α-androstane-dione and 5β-androstane-dione. In humans, 5α-androstane-dione is a metabolic precursor of both testos-terone and estrone.


4-Androstenediones is used to evaluate the function of the adrenal gland and the ovaries or the testicles as well as the production of hormones related to the development of male sex organs and physical characteristics (androgens). It is most commonly used to determine the cause of symptoms of excess androgens in a woman who has an elevated testo sterone level, along with a few other laboratory tests for this purpose.

Application

Simplified hormone drug, from the testis or urine extracted with the role of a male hormone steroids,
norethisterone, test osterone propionate in the middle Body, is widely used in small rheumatoid arthritis,
diuretic, and a variety of contraceptive to control infectious inflammation.


4-Androstenedione is the common precursor of male and female sex hormones. Some 4-Androstenedione is also secreted into the plasma, and may be converted in peripheral tissues to test osterone and estrogens.

4-Androstenedione can be synthesized in one of two ways. The primary pathway involves conversion of 17-hydroxypregnenolone to dehydroe piandrosterone by way of 17,20-lyase, with subsequent conversion of dehydroe piandrosterone to 4-Androstenedione via the enzyme 3-β-hydroxysteroid dehydrogenase.


The secondary pathway involves conversion of 17-hydroxyprogesterone, most often a precursor to cortisol, to 4-androstenedione directly by way of 17,20-lyase. Thus, 17,20-lyase is required for the synthesis of 4-androstenedione, whether immediately or one step removed.


The production of adrenal 4-Androstenedione is governed by ACTH, whereas production of gonadal 4-Androstenedione is under control by gonadotropins. In premenopausal women, the adrenal glands and ovaries each produce about half of the total 4-androstenedione(about 3 mg/day).


After menopause, 4-androstenedione production is about halved, due primarily to the reduction of the steroid secreted by the ovary. Nevertheless, 4-androstenedione is the principal steroid produced by the postmenopausal ovary.


4-Androstenedione is further converted to either test osterone or estrogen.


In males, conversion of androstenedione to test osterone requires the enzyme 17β-hydroxysteroid dehydrogenase.


In females, androstenedione is released into the blood by theca cells. Conversion of androstenedione to estrogen (e.g., estrone and estradiol) requires the enzyme aromatase. Androstenedione is a substrate for estrogen production in granulosa cells which produce aromatase. Thus, theca cells and granulosa cells work together to form estrogens.

Effects


Because androstenedione can be converted to estrogenic steroids, people taking this supplement may have estrogenic side-effects.


4-Androstenedione has been shown to increase serum test osterone levels over an eight-hour period in men when taken as a single oral dose of 300 mg per day, but a dose of 100 mg had no significant effect on serum test osterone.


However, serum levels of estradiol increased following both the 100 mg and 300 mg doses. The study also reported that the serum level of estrogens and test osterone produced varied widely between individuals.


In child development, higher levels of androstenedione in boys have been associated with higher levels of acting out behaviors.

Evaluate adrenal gland function and to distinguish between androgen-secreting conditions that are caused by the adrenal glands from those that originate in the ovaries or testicles, if results of DHEAS and test osterone testing are abnormal

Help diagnose tumors in the outer layer (cortex) of the adrenal gland or tumors outside of the adrenal gland that secrete ACTH (ectopic) and separate these conditions from ovarian or testicular tumors and cancers

Diagnose congenital adrenal hyperplasia (CAH) and monitor CAH treatment, in addition to tests for test osterone and 17-hydroxyprogesterone, for example

Help diagnose polycystic ovarian syndrome (PCOS) and help rule out other causes of infertility, no monthly menstrual periods (amenorrhea), and excess body and facial hair (hirsutism) in women who have abnormal results on tests for DHEAS, test osterone, and other hormones such as FSH, LH, prolactin, and estrogen

Investigate and diagnose the cause of male physical characteristics (virilization) in young girls and early (precocious) puberty in young boys

Help determine the cause of delayed puberty and investigate suspected ovarian or testicular failure
